Modes of uptake of methotrexate by normal and leukemic human leukocytes in vitro and their relation to drug response.

چکیده
Characteristics of Methotrexate transport by normal and leukemic human leukocytes were measured in vitro. At high external drug levels (3:20 MM) drug uptake by all cell types was temperature-insensitive, nonsaturable, and nonconcentrative. The steady-state cell/medium drug distribution ratio was about 0.03. During clinical treatment with Methotrexate, plasma drug levels generally reach 0.2 MM. At this lower drug level: (a) A temperature-sensitive and saturable mode of transport was demonstrable in all cell types except small lymphocytes. In normal granulocytes the steady-state drug distribution ratio reached 0.2. (b) Leukemic cell types showed a wide variation in capacity for uptake of the drug, and an association was seen between this capacity and clinical response to the drug.

متن کاملHepatorenal Protective Effects of Sesame Seeds Oil, Flaxseed Oil and their Mixture against Methotrexate Toxicity in Rats
Background: Methotrexate (MTX) is an anti-metabolite drug used in the treatment of many cancers and autoimmune diseases. Methods: This study investigated the protective effect of flaxseed oil, sesame seed oil, and their mixture on the MTX-induced hepatorenal toxicity. Thirty rats divided into five groups of: normal control, MTX control, and flaxseed oil, sesame seed oil, and the mixture groups...
متن کاملPreparation of Methotrexate loaded PLGA nanoparticles coated with PVA and Poloxamer188
Objective(s): Nanoparticles offer an attractive platform for drug delivery through a wide variety of the body's physiological barriers. Furthermore, modification of nanoparticle surface with moeites such as Poloxamer188 can enhance their accumulation and localization at disease site. In this work, we investigated the physiochemical effect of a scavenger receptor (SR-BI) interac...
متن کاملInvestigation of the Effect of Folic Acid Based Iron Oxide Nanoparticles on Human Leukemic CCRF-CEM Cell Line
Abstract Background Nanoparticulate drug delivery systems have attracted significant attention in the field of cancer nanotechnology. This study determines the effect of folate-based Fe2O3 nanoparticles. This study aimed to decorate nanoparticles with folate (FA), a molecular ligand for ‘active’ targeting of cancerous cells and the application of modified-nanoparticles in cancer treatment. ...
متن کاملEffect of Propranolol on Angiogenic Factors in Human Hematopoietic Cell Lines in vitro
Background: Beta-adrenergic blocking agents have been broadly used for treatment of many cardiovascular diseases such as arterial hypertension and ischemic heart failure. Anti-tumoral, anti-inflammatory and anti-angiogenesis effects of propranolol (a non-selective beta-adrenergic blocker) have been shown. Angiogenesis (replenish of the pre-existing vascular networks) plays a critical role in s...
